Semaglutide is a long-acting GLP-1 receptor agonist. It increases glucose-dependent insulin release, suppresses glucagon, slows gastric emptying and reduces appetite via hypothalamic signalling; STEP-1 (NEJM 2021) reported mean weight reduction of 14.9% over 68 weeks.
